Abstract:
While Gateway® Cloning has been popularized for its efficiency in transferring genes into various vector systems, it lacks a standardized protocol for simultaneously cloning pooled libraries. Additionally, while the DNASU Plasmid Repository has a collection of Gateway® Entry Vectors, containing over 18,000 open reading frames (ORF) of human genes, these alone do not provide enough information on gene behavior and cannot be readily used in therapeutics. One of our lab’s goals is to make a membrane display platform of the human ORFeome, which can be used for numerous research applications, such as drug screening assays and antibody discovery. Thus, this project focuses on optimizing the pooled Gateway® LR reaction to make a library of expression vectors hosting genes found in the human ORFeome.
